Application of human proteome chip in discovering new gastric cancer serum markers

According to WTO statistics, in 2012, there were 952,000 new cases of gastric cancer and 723,000 deaths. The morbidity and mortality rate were the second and third most common malignant tumors. In China, 3 people died of gastric cancer every minute. According to the statistics of the survival rate of gastric cancer patients in Ruijin Hospital in the past 30 years, the 5-year survival rate after early treatment is more than 90%, and the late stage is less than 10%. Therefore, the early diagnosis of gastric cancer has great practical significance. At present, the specificity and sensitivity of commonly used gastric cancer tumor markers such as carcinoembryonic antigen (CEA) are not high, so it is urgent to adopt a new technology platform to screen gastric cancer diagnostic markers.

Protein chips are characterized by high throughput and parallel analysis, and are particularly suitable for finding serum markers of various immune-related diseases including tumors. In the study, the researchers used gastric human microarray containing about 17,000 proteins to analyze the serum of 37 gastric cancer patients and 50 healthy people, and found 44 differentially expressed autoantibodies. Then use the protein corresponding to the 44 antibodies to customize the chip and expand the sample size for the second round of screening. In the experiment, 300 cases of gastric cancer, 314 cases of high risk of gastric cancer, and 300 healthy human serum were selected, and finally 7 candidate biomarkers were obtained. Then, sera of 300 cases of gastric cancer and healthy people were used, and ELISA was performed by taking COPS2 and CTSF as examples. The authors selected the tissues of gastric cancer and other digestive tract tumors for immunohistochemical analysis, and found that the expression of CTSF in gastric cancer tissues was significantly higher than that of other groups, and it was specific for gastric cancer. Based on the correlation analysis between markers and survival rate of gastric cancer patients, the 5-year survival rate of patients with high-grade patients was significantly higher than that of patients with low expression. The sensitivity, specificity and accuracy of the seven gastric cancer serum markers selected for the diagnosis of gastric cancer were significantly higher than the currently used single or multiple traditional markers (CEA, CA25). , CA72-4, CA9), showing high clinical application value of gastric cancer screening and early diagnosis.

The proteomic chip-based serum marker discovery process established in this study is versatile and can be easily transplanted to systematic findings of serum biomarkers of other tumors and immune-related diseases. The serum biomarkers found have significant industrialization potential in early diagnosis and prognosis of gastric cancer. The research has obtained 7 authorized patents, and is actively cooperating with relevant enterprises to promote the transformation and application of this achievement, and hopes to benefit patients with gastric cancer as soon as possible.
